Ilea Contractility Potential of Hydro-Ethanolic Extract of Oyster Mushroom ( Pleurotus Ostreatus ) on Guinea Pig ( Cavia Porcellus ) Ileum
Abstract
Ileum of animals demonstrate responses to contractile potentials of acetylcholine and its agonist herbal extracts in an isolated tissue experiment. Contractile potentials of hydro-methanolic extract of oyster mushroom (Pleurotus ostreatus) on isolated ileum of guinea pig (Cavia Porcellus) was studied. Ten milligram (10mg) of acetylcholine was serially diluted to obtain a concentration of 0.1ug/ml, 1ug/ml, 10ug/ml, 100ug/ml and 1mg/ml. P. ostreatus hydro- methanolic extract was also diluted from a stock solution of 200mg/ml to obtain a serial dilution of 20ug/ml, 200ug/ml, 2mg/ml, and 20mg/ml. Tyrode solution was used as Physiological solution for the study. Guinea pig ileum cut at 10cm from the ileocecal junction was use for the contractility study. Different concentrations of acetylcholine were used as single administrations and in combination with Pleurotus ostreatus. This was used to determine the effects of Pleurotus ostreatus hydro-ethanolic extracts on the enteric acetylcholine receptors, the results showed that the P. ostreatus extract demonstrates a physiological agonist potential to acetylcholine. It expressed contraction inducement similar to acetylcholine-induced contraction on the invitro ileum contractility test without necessarily occupying the same receptor.
