Bacteriological Analysis of Fruits Consumed in Rivers State University Female Hostels

Abstract

Bacteriological studies were conducted on a variety of fruits consumed by residents of the female hostels at Rivers State University. A total of twenty samples each of five different types of fruits—watermelon, apple, banana, and orange—were collected from vendors and hawkers near the NDDC hostel, hostel H, hostel C, and hostel B. These samples were surface- sterilized using distilled water and analyzed following standard microbiological procedures. The homogenized samples were cultured on bacteriological media using the spread plate technique and then incubated aerobically at suitable temperatures and times. Pure cultures of the resulting microbial isolates were obtained from subcultures of the primary plates. These isolates were identified through morphological, microscopic (using Gram staining and lactophenol cotton blue staining), and biochemical methods. The investigation revealed that the mean Total Heterotrophic Bacterial Count (THBC) ranged between 2.2 x 105 cfu/ml and 4.6 x 105 cfu/ml. The results also showed a Total Coliform Count (TCC) between 1.5 x 102 cfu/ml and 3.1 x 102 cfu/ml, a Faecal Coliform Count (FCC) from 1.8 x 101 cfu/ml to 3.9 x 101 cfu/ml, and a Total Staphylococcus Count (TSC) ranging from 11.1 x 104 cfu/ml to 3.4 x 104 cfu/ml. Additionally, the Total Salmonella-Shigella Count (TSSC) varied from 2.3 x 103 cfu/ml to 4.3 x 103 cfu/ml, while the Total Vibrio Count (TVC) ranged from 2.0 x 102 cfu/ml to 3.5 x 102 cfu/ml. In total, 69 microbial isolates belonging to 25 different genera were identified, including Bacillus spp., Pseudomonas spp., Escherichia coli, Staphylococcus aureus, Salmonella spp., Enterococcus spp., and others. While the presence of these bacteria and fungi indicates a need for improved sanitary practices among fruit vendors, it also presents an opportunity for growth.
